Southern Blotting
DNA molecules that have been separated by
gel electrophoresis can be identified by
hybridization with a radiolabeled probe
DNA is transferred from the gel to a
nitrocellulose or nylon filter
The DNA on the membrane is denatured, and
the filter is incubated with the probe under
controlled conditions of salt and temperature
The filter is washed to remove unbound
probe, dried, and exposed to X-ray film
